ERN1 (IRE1)/XBP1-mediated signalling plays roles in the coordination of metabolic and immune responses by acting as a regulatory hub, linking endoplasmic reticulum homeostasis with innate immunity and metabolism.
Hyperactivated ERN1 (IRE1α) increases TXNIP mRNA stability by reducing levels of a TXNIP destabilizing microRNA, miR-17. In turn, elevated TXNIP protein activates the NLRP3 inflammasome, causing procaspase-1 cleavage and interleukin 1β (IL-1β) secretion.
ERN1 binds to cholera toxin in the endoplasmic reticulum to activate RIG-I innate immune signalling.
