VENTX plays a pivotal role in human macrophage terminal differentiation and proinflammatory function. VENTX is upregulated during monocyte to macrophage differentiation, and ablation of VENTX in monocytes profoundly impairs their differentiation to macrophages.

The encoded protein may function as a transcriptional repressor and be involved in mesodermal patterning and hemopoietic stem cell maintenance. Multiple pseudogenes exist for this gene.
