Summary |
IKBKE interacts with NKFB2 (p52) and promotes transactivation via RELA (p65)-regulation of the alternative NF-kappaB activation pathway involving p52 and p65.
IKBKE is an IkappaB kinase that is expressed mainly in immune cells, and is induced in response to pro-inflammatory cytokines such as tumour necrosis factor (TNF)-alpha, IL-1-beta and IL-6, in addition to LPS.
IKBKE and TBK1 have a pivotal role in coordinating the activation of IRF3 and NF-kappaB in the innate immune response.
IKBKE and other IKK kinases regulate each other by an intricate network involving phosphorylation of their catalytic and regulatory (NEMO, TANK) subunits to balance their activities during innate immunity.
IKBKE kinase modulates IL-17 signalling through TRAF3IP2 adaptor protein phosphorylation, resulting in the induction of neutrophilia and pulmonary inflammation. (Demonstrated in mouse)
IKBKE is an important signalling molecule in the IFN pathway and is necessary to mount anti-viral immunity against West Nile virus. (Demonstrated in mice)
IKBKE plays a critical role in regulating the balance between the type I and type II interferon (IFN) signalling pathways.
IKBKE is sequestered by arenavirus nucleoproteins to block its autocatalytic activity and its ability to active IRF3.
IKBKE is poly-ubiquitinated at Lys30 and 401 to promote NF-kB activation in macrophages stimulated with TNFA and IL1B.

Summary |
IKBKE is a noncanonical I-kappa-B (see MIM 164008) kinase (IKK) that is essential for regulating antiviral signaling pathways. IKBKE has also been identified as a breast cancer (MIM 114480) oncogene and is amplified and overexpressed in over 30%% of breast carcinomas and breast cancer cell lines (Hutti et al., 2009 [PubMed 19481526]).[supplied by OMIM, Oct 2009] |
