Summary
MAP3K8 negatively regulates interferon-beta production in macrophages and myeloid dendritic cells.
MAP3K8 is essential for IL-1beta production from both macrophages and dendritic cells and an important mediator for collaboration of pattern recognition receptors with danger-associated molecular patterns (DAMPs) to induce TNF and IL-1beta production and optimal host defence.
MAP3K8 regulates IL23A expression in lipopolysaccharide (LPS)-stimulated macrophages through extracellular signal-regulated kinase (ERK) activation.
MAP3K8 is a MEK kinase that is require for the activation of MAP kinases in myeloid cells following TLR and TNF receptor stimulation. MAP3K8 is critical for production of the pro-inflammatory cytokine TNF during inflammatory responses.

Summary
[Mus musculus] Map3k8 is a MEK kinase that is require for the activation of MAP kinases in myeloid cells following TLR and TNF receptor stimulation. Map3k8 is critical for production of the pro-inflammatory cytokine TNF during inflammatory responses.
[Mus musculus] Map3k8 plays a role in facilitating both innate and adaptive antiviral responses by transducing Type I interferon (IFN) signals and promoting expression of IFN-stimulated genes (ISGs).
Entrez Gene
Summary This gene is an oncogene that encodes a member of the serine/threonine protein kinase family. The encoded protein localizes to the cytoplasm and can activate both the MAP kinase and JNK kinase pathways. This protein was shown to activate IkappaB kinases, and thus induce the nuclear production of NF-kappaB. This protein was also found to promote the production of TNF-alpha and IL-2 during T lymphocyte activation. This gene may also utilize a downstream in-frame translation start codon, and thus produce an isoform containing a shorter N-terminus. The shorter isoform has been shown to display weaker transforming activity. Alternate splicing results in multiple transcript variants that encode the same protein. [provided by RefSeq, Sep 2011]
