Summary
Hyperactivated Ern1 (IRE1α) increases Txnip mRNA stability by reducing levels of a Txnip destabilizing microRNA, miR-17. In turn, elevated Txnip protein activates the Nlrp3 inflammasome, causing procaspase-1 cleavage and interleukin 1β (IL-1β) secretion. (Demonstrated in human)
ERN1-mediated persistent reactive oxygen species generation is a mechanism used by macrophages to kill bacterial pathogens that evade the initial oxidative burst.
InnateDB Annotation from Orthologs
Summary
[Homo sapiens] ERN1 (IRE1)/XBP1-mediated signalling plays roles in the coordination of metabolic and immune responses by acting as a regulatory hub, linking endoplasmic reticulum homeostasis with innate immunity and metabolism.
[Homo sapiens] Hyperactivated ERN1 (IRE1α) increases TXNIP mRNA stability by reducing levels of a TXNIP destabilizing microRNA, miR-17. In turn, elevated TXNIP protein activates the NLRP3 inflammasome, causing procaspase-1 cleavage and interleukin 1β (IL-1β) secretion.
[Homo sapiens] ERN1 binds to cholera toxin in the endoplasmic reticulum to activate RIG-I innate immune signalling.
Entrez Gene
Summary This gene does not have any Entrez summary - the following is the summary from its human ortholog ENSG00000178607:
The protein encoded by this gene is the ER to nucleus signalling 1 protein, a human homologue of the yeast Ire1 gene product. This protein possesses intrinsic kinase activity and an endoribonuclease activity and it is important in altering gene expression as a response to endoplasmic reticulum-based stress signals.
